Sustavi za izolirane organe nude brojne mogu\u0107nosti u bazi\u010dnim farmakolo\u0161kim, fiziolo\u0161kim i patofiziolo\u0161kim istra\u017eivanjim kardiovaskularnog sustava.<\/p><p>Oscar Langendorff\u00a0 je 1897. godine opisao tehniku izolacije srca koje i izvan tijela nastavlja sa svojom aktivno\u0161\u0107u i pri tome je mogu\u0107e pratiti razli\u010dite parametre sr\u010dane funkcije. Model se zasniva na retrogradnoj perfuziji srca hranjivom otopinom kroz aortu i ostia coronaria u koronarne krvne \u017eile, \u0161to omogu\u0107uje du\u017ee izvantjelesno o\u010duvanje srca. Nepretkom tehnike sustav je modificiran i unaprije\u0111en, ali je izvorni princip funkcioniranja ostao nepromijenjen. Sustav za izolirano srce \u201eprema Langendorffu\u201c danas je standardan model u temeljnim farmakolo\u0161kim istra\u017eivanjima. Primjenjuje se za in vitro\u00a0 istra\u017eivanja izravnih u\u010dinaka razli\u010ditih tvari na parametre sr\u010dane funkcije u odsutnosti neuralnih i humoralnih \u010dimbenika. To zna\u010di da je svaka promjena sr\u010dane aktivnosti izravni u\u010dinak tvari koju primjenjujemo i nije \u201emaskirana\u201c kompenzatornim mehanizmima intaktnog organizma. Na sustavu za izolirano srce mogu se istra\u017eivati u\u010dinci razli\u010ditih tvari u bazalnim i uvjetima ishemije i reperfuzije. Uvjeti ishemije-reperfuzije vjerno opona\u0161aju uvjete infarkta miokarda pa se na njemu mogu tako\u0111er istra\u017eivati supstance koje su potencijalni kardioprotektori tijekom ishemije i\u00a0 reperfuzije. Izolirane krvne \u017eile (aorta, mezenterijalne arterije, koronarne arterije) se uzmu od pokusne \u017eivotinje i nare\u017eu na prstenove veli\u010dine 3-4 mm. Izolirani prstenovi se razapnu izme\u0111u dvije \u017eice, jedna je fiksirana za organski bazen\u010di\u0107, a druga je spojena sa pretvara\u010dem sile tlaka. Prstenovi se napregnu silom od 20 mN, stabiliziraju kroz 60 minuta, a nakon toga se prekontrahiraju noradrenalinom. Nakon \u0161to je postignut plato kontrakcije dodaje se acetilkolin koji slu\u017ei za procjenu o\u010duvanosti endotela krvne \u017eile. Relaksacija na acetilkolin mora biti ve\u0107e od 50% da bi \u017eile smatrali funkcionalnim. Nakon toga slijedi izvo\u0111enje planiranog pokusa i istra\u017eivanja potencijalno vazoaktivne supstance.<\/p><p>Srce i krvne \u017eile uzimaju se od pokusnih \u017eivotinja kao \u0161to su mi\u0161evi, \u0161takori, zamor\u010di\u0107i ili kuni\u0107i. Izolirani organi se u in vitro uvjetima prehranjuju Krebs-Hensseleitovom otopinom koja je oksigenirana i zagrijana na temperaturu od 36,7 C.<\/p><p>Osim u istra\u017eiva\u010dke svrhe, sustavi za izolirane organe mogu se koristiti i u nastavne svrhe. Studenti na vje\u017ebama iz farmakologije mogu \u201eu\u017eivo\u201c vidjeti u\u010dinke brojnih lijekova na srce i krvne \u017eile. Tako\u0111er se mogu pokazivati mehanizmi dijelovanja pojedinih lijekova. Tijekom nastave iz fiziologije studenti su u mogu\u0107nosti prou\u010davati promjene na srcu prilikom promjene koncentracije kisika, hranjivih tvari ili elektrolita u otopini kojom se hrani srce.<\/p><p>Ovi sustavi predstavljaju novost u Mostaru i Bosni i Hercegovini te daju novu dimenziju znanstveno-istra\u017eiva\u010dkom radu na na\u0161em fakultetu. Tako\u0111er pru\u017eaju mogu\u0107nost studentima za izradu diplomskih radova i lije\u010dnicima za izvo\u0111enje pokusa za svoje magisterije i
